How does reference-standard error propagate into an orforglipron content figure?

I am comparing a supplier certificate against an independent result on the same lot.

The units are where I keep going wrong, so please be explicit about them.

I have sanity-checked the order of magnitude and it seems right, which is not the same as being right.

Is my approach right even if my number is wrong?

On the detail: the cost of a certified standard is cheaper than the cost of one wrong conclusion based on an unverified standard, which is the argument for using them.

Purity of the standard is not the same as traceability of the standard — purity tells you the composition, traceability tells you the standard came from a credible source.

Put another way, a standard purchased from a supplier carries a certificate stating its purity, and that purity is your starting point for understanding your result.

The pharmacopoeial standards specify system suitability requirements for quantitative methods that include replicate injection agreement and that agreement should be tighter than two per cent.

Ask for the standard's purity when you buy it.

A standard of uncertain purity used to measure an unknown sample produces a result whose uncertainty is larger than you will have estimated.

The relevant detail is that using a standard that is older than its certificate validity window is using a material of unknown purity, which defeats the purpose of having a standard.

On the detail: a standard should be stored as carefully as the samples you test it against, because a degraded standard skews all of your results in a consistent direction.

The single highest-value check is verifying that the standard's certificate specifies its purity and gives a traceable source.

The response factor changes if the method changes — a different gradient time, a different temperature, a different column — so the standard must be run under the exact method conditions as the sample.

I would not accept a standard that does not come with a certificate stating its purity.

Do not use an in-house standard without independent verification of its purity.

The response factor computed from a standard — area per unit mass — assumes the standard and sample ionise identically, which is only true if they are treated identically.

Storing a standard in a freezer is better than storing it at room temperature, but how much better depends on the chemical stability of the standard.

The caveat is that a standard certificate is only as good as the issuing laboratory is reliable, and not all certificates are equally credible.

Ask for the standard's purity when you buy it.

Most research work uses in-house standards of unknown provenance, which is exactly the situation where a mistake in the standard produces systematic error in every sample measured against it.

Most research quantitation work assumes the sample and standard are treated identically through every step, which is almost never true if they come from different sources.

One qualification: using a standard from a reputable source and checking its purity on receipt is worth the effort.

The practical summary: use a standard of declared purity and verified traceability, and check that the certificate is current.
